Interleukin 10 (IL-10) is a cytokine which is encoded for by the IL10 gene. IL-10 is a major anti-inflammatory cytokine, a protein released by cells which helps damp down the inflammatory response, and so a lack of its expression is therefore associated with chronic inflammatory disorders including Crohn’s disease, asthma, prostatitis 1 and several other disorders 2.

There is one SNP in IL10 which is associated with poor health outcomes rs1800896 or A-1117G which is associated with the development of chronic prostatitis 1.

Risk Description

The risk allele ‘A’ of A-1117G in the IL10 gene is associated with a reduction in the amount of IL-10 production. As IL-10 is a key anti-inflammatory agent this reduction is often associated with the development of inflammatory disorders such as asthma 3, Crohn’s disease 4, atopic diseases 5 and chronic prostatitis 1.

Quercetin

Quercetin is a common flavonoid molecule found in many foods with reddy or dark green vegetables being a particularly rich source 6.

Quercetin exhibits a potent inhibitory effect on the immune cells and so will damp down the pro-inflammatory environment. In those carrying the risk ‘A’ allele of A-1117G, which is associated with a reduced anti-inflammatory character, quercetin may therefore be beneficial in limiting inflammation and associated symptoms 1.

Interestingly, those with the “normal” ‘G’ allele of rs1800896, who also carried the risk ‘A’ allele of rs1800629 in the TNFA gene responded poorly to quercetin treatment, which may explain some of the variation observed for this therapy.
