FGFR1
FGFR1 is a receptor protein expressed in a very broad array of tissues, where it regulates a wide range of processes, including cell division, differentiation and metabolism. Its many roles include co-operation with the anti-aging and neuroprotective protein Klotho in the kidney 1. These proteins co-recruit a type of fibroblast growth factor known as FGF23, which regulates phosphate and calcium homeostasis 2.

Risk Description
This variant does not affect the amino acid composition of the FGFR1 protein, but positively affects its expression. The minor C allele of this variant is associated with higher FGFR1 expression. The same allele also shows a strong association with lower levels of the circulating Klotho protein 3, which is thought to confer anti-aging and neuroprotective benefits 4, 5, 6. The mechanism of this effect may involve a competition between the FGFR1-bound and circulating Klotho.
