MAP kinase-activating death domain protein (MADD) is an adaptor protein encoded for by the MADD gene. MADD interacts with another protein TNF-alpha receptor 1 to activate mitogen-activated protein kinase signaling within cells, in response to apoptotic (controlled cell death) signals, enabling the cell to die in a controlled fashion1.  There is evidence that the SNP rs7944584 is associated with increased circulating glucose levels in man, although a mechanism remains undescribed2.

Risk Description

The risk allele ‘T’ of rs7944584 in the MADD gene has been associated with a decreased conversion of pro-insulin to insulin which leads to higher circulating glucose levels. Interestingly however the risk allele was also associated with markers of improved clinical markers of insulin sensitivity in the same study2,3.
